Chemical structures of the sugar chains of human transferrin (Tf) from patients with hepatocellular carcinoma (HCC) and from liver cirrhosis were examined by pyridylamination of their oligosaccharides in combination with reversed-phase and size-fractionation high-performance liquid chromatography, and were compared with those of human alpha-fetoprotein (AFP). Crossed immunoaffinoelectrophoresis of Tf was performed in the presence of concanavalin A (Con A) and Lens culinaris agglutinin (LCA). Two, reactive and nonreactive, and 3 peaks, reactive, weakly reactive and nonreactive, were detected on crossed immuno-affinoelectrophoresis in gels containing LCA and Con A, respectively. LCA-reactive species of Tf from HCC was higher than that from LC with statistical significance, and the carbohydrate analysis showed that the ratio of the fucosylation at the innermost N-acetylglucosamine residue on the biantennary sugar chain in HCC was higher than that in liver cirrhosis. Carbohydrate analysis of AFP in HCC also showed an increased ratio of the fucosylation at the innermost N-acetylglucosamine residue in biantennary sugar chain. However, there was no statistical significant correlation in the ratios of fucosylation on the biantennary sugar chain between Tf and AFP. On the other hand, the percentage of Con A-nonreactive plus-weakly reactive species in HCC was statistically higher than that in LC. Positive correlation was observed between the percentage of Con A-nonreactive pulse-weakly reactive species and the ratio of the fucosylation in biantennary sugar chain of Tf. Thus, these results indicate that the increment in the fucosylation and branching to form new antennae is a characteristic features of the carbohydrate chains of Tf in patients with HCC.
