Experimental liver damage induced by lipopolysaccharide (LPS) administration following Propionibacterium acnes (P. acnes) injection were studied in rat.<BR>Multiple granulomas in liver and numerous splenic mononuclear cells (SMNC) at the margenal zone and red palp were observed 7 days after injection of P. acnes in rats (P-rat). Following injection of LPS induced massive hepatocytes necrosis.<BR>Splenectomy before the administration of LPS reduced the degree of liver damage and the number of the infiltrating cells in granulomas.<BR>The injection of the splenic adherent cells of the P-rat with LPS via portal vein induced severe liver damage in P. acnes pretreated rat.<BR>PHA induced blastogenic activities of SMNC was reduced significantly after the injection of P. acnes and LPS. Killer activities of SMNC could not be detected through the experimental periods.<BR>The culture supernatant of P-rat splenic adherent cells with LPS, showed the cytotoxic effect on normal isolated hepatocytes.<BR>These results indicated SMNC, especially adherent cells, played an important role in the induction of hepatocytotoxicity on this experimental model.
