Metabolism of 3β, 7β-dihydroxy-5β-cholan-24-oic acid in man

Abstract

Analysis of 3β, 7β-dihydroxy-5β-cholan-24-oic acid (3β, 7β-diOH), which was found in serum from gallstone patients treated with ursodeoxycholic acid (UDCA) and identified for the first time by us, in serum, bile and urine was carried out. This bile acid was detected in serum from the fasting healthy subjects and the value was 0.08±0.07μg/ml(mean±SD, n=5).<BR>To determine the ratio between free and amino acid conjugated bile acid in serum, the analysis was performed excluding the conjugation procedure in 4 healthy subjects and 3 gallstone patients administered with UDCA for seven days, and the serum level of individual bile acid was compared with the level (free plus conjugated bile acid) determinned by the complete method of gas-liquid chromatography. The ratios of 3β, 7β-diOH to the free plus conjugated bile acid were around 1.O in all subjects. This result indicates that 3β, 7β-diOH exists in blood as unconjugated form.<BR>Bile was collected from 14 gallstone patients treated with UDCA. 3β, 7β-diOH could not be detected in any trace amount in the bile, suggesting that the bile acid was not excreted into bile.<BR>The urinary excretion of this bile acid was observed in 3 gallstone patients. In two the bile acid in urine was unconjugated form, while in one the bile acid occured as amino acid conjugated form. Sulfated form was detected in trace amount. Urinary clearance of 3β, 7β-diOH was lower than that of other bile acids.
