An attempt was made to develop a rapid quantitative determination method of a micro amount of medicinal drugs by atomic absorption spectroscopy. According to the metal complex formation of medicinal drugs, they may be classified into several groups, and cyanocobalamin, vitamin B12, belongs to the group which contains cobalt, i.e., a drug itself is a metal chelate compound. Therefore, it is very natural to consider that the determination of vitamin B12 can be achieved by measuring cobalt contents by atomic absorption spectroscopy. A Hitachi Model 207 atomic absorption spectrophotometer, equipped with a premix type burner, using air and acetylene fuels and a hollowcathode lamp was used throughout the measurements.<BR>In this paper, determination conditions were mainly investigated. For a comparison, α-cobaltic glycinate, synthesized in our laboratory, as well as cobaltous acetate purchased were employed. Both compounds are water soluble and acetate is also alcohol soluble, while cobaltic glycinate being insoluble in alcohol. It is very convenient to measure known compounds simultaneously, for the establishment of the measuring conditions of cyanocobalamin. The operating conditions are as follows: wavelength, 2407 Å; lamp current, 10 mA ; slit width, 0.18 mm; burner position, the lowest position; and flow rate of air, 13.0 l/min and of acetylene, 3.0 l/min.<BR>The results show that the recovery of cobaltous acetate and cobaltic glycinate obtained in water solution was unexpectedly low, however, when measurements were made in 0.1 N HCl solution, it gave nearly 100% recovery. Then cyanocobalamin, as well as its preparations, such as injections and tablets, are measured in 0.1 N HCl solution. In alcoholic solution, fuel conditions should be readjusted, depending on the mixing ratio of the solvents. In 20% C2H5OH solution, cyanocobalamin gave low values, while in 100% C2H5OH, cyanocobalamin and hydroxocobalamin gave an expected value. In 0.1 N HCl C2H5OH solution, it gave also better values.<BR>In conclusion, determination of cyanocobalamin preparations are recommended to be carried out in 0.1 N HCl solution, even when they are water soluble. Alcohol solution is also available for this measurement.<BR>As injections and even tablets are easily soluble in both water and alcohol, they are measured with great ease and therefore, this method is recommended to be employed as a rapid quantitative determination method pharmaceutical industries.<BR>There are many types of structures in medicinal drugs, when classified according to the metal complex formation, and determination of drugs by atomic absorption spectroscopy will be capable of measuring even a micro amount of metals in drugs, when they would form metal complexes.<BR>The authors are attempting to develop this method to the determination of medicinal drugs in biological materials.
