In order to know whether DNA methylation is involved in the regulation of human pepsinogen gene expression, we have analyzed the expression and methylation of human pepsinogen A (PGA) genes, the main component of human pepsinogen genes, in various human tissues. An expression of PGA mRNA of about 1.8kb in size was limited only to the fundic mucosa of the stomach and not observed in any other tissue. In the fundic mucosa of the stomach, both 5' and 3' region of PGA genes were hypomethylated relatively more than in any other tissue. Thus, PGA production was regulated at least at the level of its mRNA expression and the mechanism utilizing DNA methylation was suggested in the regulation of PGA genes expression.
