Preparation of a murine transfer factor (TFd) and its biochemical properties were investigated. TFd was prepared from the homogenates of non-adherent spleen cells of mice primed with either sheep red blood cells or methylated bovine serum albumin. A passive transfer of the delayed type hypersensitivity (DTH) reaction to unprimed recipient mice became possible by subcutaneous injection with such amount of TFd as equivalent to 1×107 primed cells.The degree of DTH reaction in the recipients of TFd, however, was lower than that in those given the primed spleen cells.<br>TFd was divided into five fractions (Fr 1 to Fr 5) by gel filtration with Sephadex G-25. The third peak (Fr 3) possessed the most potent activity. Furthermore, it was revealed by thin-layer chromatography with PEI-cellulose that Fr 3 consisted of at least two components. Active component (s) in TFd with a molecular weight of less than 3, 500 daltons could be inactivated partially by treatment with nuclease Pl and completely by heating at 90C for 15 min, but not at all by treatment with DNase, RNase or protease.
