The present communication has four main experiments.
1. The effect of various nucleotides and D-ribose on the maintenance of motility of chicken sperm diluted with RINGER'S solution in vitro was studied. A single addition of inosine-5'-monophosphoric acid disodium salt, cytidine-5'-monophosphoric acid disodium salt, uridine-5'-monophosphoric acid disodium salt and D-ribose to the semen diluents did not show any positive response. Guanosine-5'-monophosphoric acid disodium salt, adenosine-5'-monophoric acid, adenosine-5'-triphosphoric acid disodium salt and thymidine-5'-monophosphoric acid inhibited the motility when their hypertonic solution was used.
2. In in vitro experiments, a incorporation of inosine and its related compounds into chicken sperm was studied using 14C-labeled compounds. The incorporation of inosine-8-14C, adenine-8-14C and hypoxanthine-8-14C into the chicken sperm was confirmed by microautoradiograpy.
3. The fertility of eggs collected after the artificial insemination in chicken using the semen diluted with RINGER'S solution containing inosine, adenine or inosine-adenine mixture was examined. A higher fertility was recognized in a single addition of inosine, while the adenine did not show any positive effect. The mixture of inosine and adenine lowered the fertility.
4. The effect of inosine orally supplied on the production of chicken sperm was investigated during for 77 days, and it was found that the number of sperm per ejaculate and their motility increased.
