Abstract

Objective: The diversity of myoepithelial cells of pleomorphic adenoma in salivary gland was confirmed by ultrastructural and immunohistochemical studies.<BR>Study Design: 5 cases of pleomorphic adenoma diagnosed by cytology and histology were searched in ultrastructural observation and immunohistochemical staining (cytokeratin, GFAP, vimentin, S-100 protein and SMA).<BR>Results: (1) Cells constituting a pleomorphic adenoma were classified into ductal and other cells. Ductal cell features resemble intercalated intercalated duct cells of the normal salivary gland, while other cells resembled normal myoepithelial cells.<BR>(2) Other cells showed changes, and round to oval cells with several desmosomes and weakly positive for cytokeratin were epithelial.(3) Spindle to squamous metaplastic cells showed normal myoepithelial cell features.(4) Asteroid to small round single cells had pinocytotic vesicles, glycogen, and proteoglycan granules in cytoplasm, strong positivity for GFAP, vimentin, and mesenchymal S-100 protein and resembled chondroid metaplastic cells.(5) Focal cells of ductal cells were observed to be the reserve cells rich in glycogen and intermediate filaments in cytoplasm.<BR>Conclusion: The occurrence of this tumor was considered most intercalated duct cells in the salivary gland. Diverse cell changes were modified myoepithelial cells that showed morphological features in spindle, round, and chondroid metaplastic cells, and cells showed a diversity of character.
