A genomic DNA for the vitellin-degrading protease (Vtn protease) was isolated from a genomic library of Bombyx mori and sequenced. The Vtn protease gene was organized with 5 exons interrupted by 4 introns. This exon/intron organization was rigidly conserved in mammalian trypsin genes, but was not found in insect trypsin genes so far sequenced. Vtn protease gene was present as a single copy in the haploid B. mori genome. A cluster of the concensus sequence, designated as a Dfd binding site, a homeodomain protein binding site, was observed in the 5′ flanking region, suggesting the involvement of homeotic proteins in the regulation of Vtn protease gene expression.
