Cross-linked polystyrene containing nucleic acid bases such as uracil (U) or thymine (T) was prepared. The purpose was to separate adenosine (A) from guanosine (G) in mixed solutions of A and G by adsorption chromatography. The adsorption isotherms of A proved that A is chemically adsorped in the resin. The experimental conditions which show higher selectivity for A were surveyed by column method. No effect of space velocity was found. The resins of lower degree of cross-linking, in which the content of U is higher, showed higher amounts of adsorption of nucleoside per content of U in the resin; the resin of higher degree of cross-linking, however, showed higher selectivity for A. In the case of lower cross-linked resin, a higher relative ratio of adsorption (A/G) was obtained as the concentration of (A+G) solution decreases. In the case of higher cross-linked resin, a higher adsorption ratio (A/G) was obtained as the concentration of (A+G) increases. The fact that A was more adsorped than G in all cases suggests that A is hydrogen-bonded to U or T in the resin to form a base pair.
