Collagen prepared from arrow squid mantle muscle was immersed in a bacterial collagenase solution at several concentrations (enzyme : substrate = 1 : 10, 1 : 100 and 1 : 1, 000), and the reaction mixture was analyzed by SDS-PAGE. Collagen degradation was increased with increasing collagenase concentration. This result suggested that long-term exposure to a collagenase preparation could degrade collagen, even at a low reaction temperature (5°C). The mantle muscle of arrow squid was immersedin a collagenase solution at several concentrations (0.1, 1 and 10 mg/ml), and the effect of the collagenase was studied in comparison with that of the control solution without collagenase. The shear force of each tested sample was significantly lower than that of the control. Light microscopy detected voids within the muscle at the earliest time in the 10 mg/ml solution. Transmission electron microscopy clarified that these voids within the muscle already existed after only 3 h in the collagenase preparation. These results suggest that the collagen degradation was promoted by the collagenase preparation and that collagen degradation might have been responsible for the softening of the squid muscle during refrigeration.
