Non-hibernating lethal egg (l-n) was discovered by Drs. Hirobe and Ooi 1951. When the heterozygous individuals (l-n/+) of this strain were crossed mutually, the normal eggs and the lethal ones (l-n/l-n) were oviposited at the ratio 3:1 in the same back. The embryos of these lethal eggs develop until blastokinesis, whereas the embryos of normal eggs do not develop and enter diapause.
The present study was designed to make clear the developmental mechanisms of the embryo of the non-hebernating lethal egg. The results obtained are as follows;
1) The expresion of lethal gene is recognized on some tissues in the egg of l-n/l-n individual by histological observation. The expression appears in serosal cells and yolk nucleus at very early stage, but it appears considerably late in the embryo of lethal egg. The pigments in serosal cells cannot be formed but the embryo develops. From these observations, the lethal egg seems to be non-hiternating one.
2) As shown in the photographs, remarkable differences of histochemical reaction between the normal and the lethal eggs are recognized on the yolk nuclei and their surrounding meshy tissues. The nuclei are gradually destructed and the meshy tissues are not formed by the action of l-n gene.
3) From the genetical consideration and histological observation, it can be presumed that the yolk nucleus of the normal egg (diapause) has an important effect on the embryonic development, because it is evident that the yolk nucleus gives an information to produce some substances inhibiting the embryonic development in the ooplasm. But in the case of l-n/l-n eggs, the inhibiting substances cannot be formed due to the destruction of yolk nuclei, and then the embryo of l-n strain can develop.
