抄録

The purpose of the present study is to investigate the action site of exfoliative toxin (ET) in the epidermis by immuno-electron microscopy using protein-A colloidal gold complex method. After S. aureus strain ZM was cultured, ET was separated by modified Kondo's method. Anti-ET serum was obtained by immunization of rabbis with purified ET mixed with Freund complete adjuvant. Purified ET was applied to the normal human epidermal organ culture which was incuvated for 1.5, 2. 5 and 3.5 hours at 37°C, and specimens were prepared for light and transmission electron microscopy, immuno-electron microscopy, and indirect immunofluorescence microscopy. By light microscopy intraepidermal cleavage was observed at the level of the granular layer after 1.5 hours and extended to the upper spinous layer after 3.5 hours. By transmission electron microscopy, desmosomes were seen separated and the half-desmosome-like structure and tonofilament retracted into cytoplasm as seen in SSSS. By immunhfluorescence microscopy, intercellular fluorescence was first observed in the basal layer, and then in spinous and granular layers according to the time course. When reached the granular layer, acantholysis occurred. Acantholysis was also observed at 0°C. By immuno-electron microscopy, gold particles were mainly seen in the outer leaflet of desmosomes, but sometimes in the intercellular contact layer, attachment plaque and tonofilaments inserting insertingto desmosomes. From the present observation, it is likely that ET attaches first to the outermembrane component of desmsomes.
