抄録

1) The intramuscular administration of KW-1062 to mature rabbit at a dose of 10 mg/kg resulted in a peak serum concentration of 50. 8 gg/m1 at 30 minutes after dosing and in a peak aqueous humor concentration of 3.6 μg/ml at 1 hour after dosing.<BR>2) The subconjunctival administration of KW-1062 to mature rabbit at a dose of 15 mg (about 0.4 ml) resulted in a peak serum concentration of 30.6 μg/ml and a peak aqueous humor concentration of 19.8 μg/ml at 30 minutes after dosing. Even at 4 hours after dosing, relatively high aqueous humor cocentration of 10.3 μg/ml was observed.<BR>3) KW-1062 was injected intramuscularly to adult patients of cataract and glaucoma 1-3 hours before operation at a dose of 40 mg : most of their aqueous humor concentration were trace.<BR>4) KW-1062 was administrated to 11 patients consisting of blepharitis, phlegmone palpebrae, dacryocystitis and ulcus corneae serpens. Clinical response was good in 10 out of 11 patients, effectiveness rate being 90.9%.<BR>5) No serious side effects were observed except one case, of whom conjunctival bulbi changed degenerative after subconjunctival injection of KW-1062 (10 mg, 0.25 ml) for 10 days, but this change was reversible.
