Increased peroxidation of membrane lipid reduces the transformability of erythrocytes, eventually contributing to diabetic microangiopathy. The present study was designed to investigate the mechanism of the lipid peroxidation in human erythrocyte membrane induced by glucose. After exposing erythrocytes to a high concentration of glucose, their susceptibility to peroxidation was assayed using the H2O2-induced autoxidation method described by Stocks et al. Glycation and peroxidation were accelerated in a parallel manner at glucose concentrations (G) ranging from 5.5 mM to 55.5 mM. Erythrocyte GSH levels were also increased. Peroxidation was further enhanced by the addition of thiol compounds to the medium, and conversely, inhibited by thiol blockers. Thus, peroxidation appeared to be mediated by the reduction of metal by intracellular thiols, however, peroxidation was still enhanced by increasing G in the presence of a thiol blocker. Peroxidation was inhibited almost completely by vitamin E, and partially by both hydroxyl radical scavengers and metal chelators. These results suggest that the production of superoxide anions following glycation of hemoglobin and/or reduction of trasitional metal ions by intracellular reducing substance leads to the formation of hydroxyl radicals, thereby accelerating lipid peroxidation in the erythrocyte membrane.
