The effect of gender in thymoquinone toxicity in rats

Purpose: Thymoquinone (TQ) (2-isopropyl-5-methyl-1,4-benzoquinone) is the main constituent of the volatile oil of Nigella sativa seeds. TQ has been tested in vivo for its efficacy against several diseases such as cancer. The aim of the present study is to address the possible mechanism of toxicity using intraperitoneal (i.p.) injection of TQ and to find the Maximum Tolerated Dose (MTD) value for acute exposure of TQ in male and female rats. Methods: Thirty two male rats and an equal number of female rats were assigned into two groups; male and female. These two groups were subdivided according to the dose level under investigation: 0, 20, 30, 40 mg/kg body weight. Eight male and eight female rats were used in each dose level and were observed for five days with necropsies performed at the end of the experiment. Tissue and blood serum samples were taken for histological evaluation and biochemical analysis. Results: One-half (50%) of all male and female rats which received acute IP injection of TQ showed signs of toxicity related to acute pancreatitis. This toxicity outcome is the result of hypoglycaemic as well as hyperlipidemic effects of i.p. injection of TQ. The MTD for IP injection in females was 15 mg/kg and 22.5 mg/kg in males. Conclusion: The tolerability threshold for TQ toxicity in males is more than in females and this could be due to drug disposition differences in both genders.
