<jats:p><jats:bold><jats:italic>Background—</jats:italic></jats:bold>Infectious diseases have emerged as potential risk factors for cardiovascular disease (CVD). Epidemiological studies support a connection between periodontal disease, a chronic inflammatory disease of the supporting tissues of the teeth, and CVD.</jats:p><jats:p><jats:bold><jats:italic>Methods and Results—</jats:italic></jats:bold>To directly test the connection between periodontal disease and atherosclerosis, apoE<jats:sup>−/−</jats:sup>mice were orally challenged with the periodontal disease pathogen<jats:italic>Porphyromonas gingivalis</jats:italic>or an invasion-impaired<jats:italic>P gingivalis</jats:italic>fimbriae-deficient mutant (FimA−). Both wild-type<jats:italic>P gingivalis</jats:italic>and the FimA− mutant were detected in blood and aortic arch tissue of apoE<jats:sup>−/−</jats:sup>mice by PCR after challenge. ApoE<jats:sup>−/−</jats:sup>mice challenged with wild-type<jats:italic>P gingivalis</jats:italic>presented with increased atherosclerotic plaque and expressed the innate immune response markers Toll-like receptor (TLR)-2 and TLR-4 in aortic tissue. Despite detection of the FimA− mutant in the blood and in aortic arch tissue, apoE<jats:sup>−/−</jats:sup>mice challenged with the FimA− mutant did not present with periodontal disease, upregulation of TLRs, or accelerated atherosclerosis. Furthermore, we demonstrate that immunization to control<jats:italic>P gingivalis</jats:italic>-elicited periodontal disease concomitantly prevents<jats:italic>P gingivalis</jats:italic>-accelerated atherosclerosis.</jats:p><jats:p><jats:bold><jats:italic>Conclusions—</jats:italic></jats:bold>We conclude that invasive<jats:italic>P gingivalis</jats:italic>accelerates atherosclerosis.</jats:p>
