Experiments were carried out to analyse the mechanisms of trimolting of silkworms induced by the treatment with an imidazole compound (KK-42). The variations of the juvenile hormone (JH) titers were estimated from the morphological changes induced by ecdysterone injection. In the larvae treated at the 4th-instar, normal larval ecdysis was observed when ecdysterone was injected at 24hr after treatment with KK-42. Swollen antennae, evaginated wing discs and pupal cuticles were observed when ecdysterone was injected at 48hr and 72hr after treatment with KK-42. The morphological changes brought about by the injection of ecdysterone into the trimolters of the last instar (4th instar) which have been induced by treatment of the 3rd instar resembled those of the tetramolters of the 5th instar. Application of KK-42 to the 3rd instar larvae inhibited the increase of ecdysteroid titers for several days after the treatment and the ecdysteroid titers rose remarkedly just before the 3rd ecdysis. Based on these results, it is assumed that the JH titers decreased sharply at 48hr and reached a level of 0 at 72hr after treatment with KK-42 in the 4th instar larvae. The inhibitory effect of KK-42 on the ecdysteroid level may play an important role in the induction of trimolter by the 3rd-instar-treatment.
