Cyst of Sarcocystis sp. was isolated from a sika deer in Hokkaido (Cervus nippon ezoensis). The deer was a 3-year-old female and had been captured in Kushiro Town on 23 April 2002. The isolated cyst was spindle shaped and its size was 3mm×1mm. It had a hairy wall of 5μm thick and was interiorly divided by the septa which contained numerous bradyzoites. To identify the sample to the species level, genomic DNA was extracted for the phylogenetic analysis. The portion of 18S rRNA gene was amplified and 1,892 bp of its partial sequence was determined. The highest identity was 94.4% by the comparison of 1,534 bp sequence between the sample and S. fusiformis by a BLAST search. The identities of the sample with pathogenic S. hominis and S. suihominis were 94.2 % and 92.5%, respectively. These identities indicate that the sample is different species from the pathogenic species ever reported. Phylogenetic tree was constructed with the close related species retrieved from GenBank. The tree revealed that there was no species which composed the clade with the sample. This result suggests that the sample does not correspond to the species whose 18S rRNA sequences have been reported up to the present. In addition, there is no sequence data of Sarcocystis isolated from dee in the previous reports. Therefore, accumulation of the sequence data for various Sarcocystis species isolated from deer is needed to identify the sample to species level.
