Endothelial cells (ECs) in blood vessels adhere to each other through specialized cell-cell junctions such as adherens junctions (AJs) and tight junctions (TJs). AJs regulate not only the integrity of endothelium through EC-EC adhesion, but also the proliferation and survival of ECs, paracellular permeability to solute, and transmigration of leukocytes across endothelium. TJs play an important role in regulating paracellular permeability and maintaining cell polarity, serving as a barrier and a fence within the membrane. Confluent endothelial cells in culture are generally regarded as a model of resting endothelium in blood vessel, as they form junctions at the points of cell-cell contact, lose ability to proliferate in response to growth factors, and stay stationary. However, incompatibility between junctional integrity and endothelial cell motility remains uncertain. In this study, I try to identify the compatibility between EC adhesion and integrity by time-lapse analysis.
