Recently, many mutagenic compounds have been isolated from pyrolysates of amino acids and cooked foods. These mutagens are aromatic heterocyclic amines, and Trp-P-2 (3-amino-1-methyl-5H-pyrido [4, 3-b] indole), MeIQx (2-amino-3, 8-dimethylimidazo [4, 5-f] quinoxaline) and IQ (2-amino-3-methylimidazo [4, 5-] quinoline) are three of them. The purpose of this study was to investigate histologically the changes of epithelial cells in the tongue which was excised twice and treated with Trp-P-2 or MeIQx or IQ. In all animals in Group 1, the tongue showed only hyperkeratosis. In 3 of 5 animals in Group 2, the tongue exhibited mild epithelial dysplasia changes. In 1 of 5 animals in Group 3 and in 2 of 5 animals in Group 4, the tongue showed slight epithelial dysplasia changes. The present investigation suggested that Trp-P-2 or MeIQx or IQ treatment in combination with two excisional wounds could produce the epithelial dysplasia in the tongues of hamsters.
