Drosophila melanogaster as a model host for Staphylococcus aureus infection

<jats:p> <jats:italic>Staphylococcus aureus</jats:italic> is an important pathogen of humans, causing a range of superficial and potentially life-threatening diseases. Infection of the fruit fly <jats:italic>Drosophila melanogaster</jats:italic> with <jats:italic>S. aureus</jats:italic> results in systemic infection followed by death. Screening of defined <jats:italic>S. aureus</jats:italic> mutants for components important in pathogenesis identified <jats:italic>perR</jats:italic> and <jats:italic>pheP</jats:italic>, with fly death up to threefold slower after infection with the respective mutants compared to the wild-type. Infection of <jats:italic>D. melanogaster</jats:italic> with reporter gene fusion strains demonstrated the <jats:italic>in vivo</jats:italic> expression levels of the accessory gene regulator, <jats:italic>agr</jats:italic>, <jats:italic>α</jats:italic>-toxin, <jats:italic>hla</jats:italic>, and a manganese transporter, <jats:italic>mntA</jats:italic>. The use of the green fluorescent protein as a reporter under the control of the <jats:italic>agr</jats:italic> promoter (P3) showed <jats:italic>S. aureus</jats:italic> microcolony formation <jats:italic>in vivo</jats:italic>. The disease model also allowed the effect of antibiotic treatment on the flies to be determined. <jats:italic>D. melanogaster</jats:italic> is a genetically tractable model host for high-throughput analysis of <jats:italic>S. aureus</jats:italic> virulence determinants.</jats:p>
