The aim in putting together this 7th volume of the Bone and Mineral Research series has been to present high quality, up-to-date reviews and in-depth analyses in areas where developments are, or potentially could be, of great interest to the Bone and Mineral research community. The high standard already established in the previous six volumes of this series (under direction of Dr. William A. Peck) has been maintained. Although the regulation and mechanism of action of the major calcium regulating hormones (such as vitamin D and parathyroid hormone) have long been recognised, there are more important gaps in knowledge. This volume reviews the prevalence, patholophysiology and consequences of primary hyperparathyroidism, one of the most common endocrine disorders. It covers the potential role of vitamin D in disorders of proliferation, providing some thoughtful insights into the signals for parathyroid gland profileration and secretation, and making the important distinction between the two. The volume also contains contributions in the areas of: bone remodelling (including the cytokines and their regulation in physiology of bone call function, of bone disease; aspects of regulation of cartilage differentation and metabolism; and TGFB, one of the major non-collagenous cytokines in bone); the broad clinical classification of Sillience; and the bisphosphonates (including their use in Paget's disease, tumor induced bone disease, hyperpathyroidism and osteoporosis; and their structure, function, pharmacokinetic mechanism of action and the therapeutic implications.
