Verification of Hybrid Nature of Diploid and Triploid Progeny between Amago Salmon and Japanese Char by RAPD-PCR Analysis.

Abstract

Artificial hybridizations were made between female amago salmon, Oncorhynchus masou ishikawae, and male Japanese char, Salvelinus leucomaenis. Triploid hybrids were then produced by exposing the fertilized eggs to heat shock (27°C, for 23 min, 8 min after fertilization) . Hybrid nature and ploidy status of the resultant progeny were examined by karyotyping. The genetic constitution of the hybrids was also examined by RAPD (random amplified polymorphic DNA) -PCR (polymerase chain reaction) . Using either amago salmon or Japanese char DNA as template of 12 primers examined, five resulted in a pattern which is distinguishable between the two species. Using the hybrid DNA as template three of the five primers yielded DNA fragments from both species, suggesting their hybrid nature. These results suggest that RAPD-PCR analysis is a useful method for identification of hybrids.
