抄録

Thein vitroantibacterial activity of NM394, the active form of the prodrug NM441, was compared with that of norfloxacin (NFLX), ofloxacin (OFLX), ciprofloxacin (CPFX), tosufloxacin (TFLX) and sparfloxacin (SPFX). The results may be summarized as follows:<BR>1) NM394 demonstrated broad-spectrum antibacterial activity against gram-positive and gram-negative bacteria.<BR>2) The activity of NM394 against gram-positive clinical isolates was comparable to that of OFLX and CPFX. Against gram-negative isolates, NM394 showed antibacterial activity equal to that of CPFX, but showed the greatest activity for all compounds tested against a part of strainsEnterobacteriaceaeandPseudomonas aeruginosa.<BR>3) NM394 showed bactericidal action at the MIC against a variety of clinical isolates.<BR>4) The activity of NM394 was unaffected by inoculum size, but slightly decreased in the presence of magnesium ions.<BR>5) The antibacterial and bactericidal activity of NM394 was unaffected by human serum.<BR>6) The susceptibilities of bacteria to NM394 were slightly decreased by passage in the presence of a subMIC concentration of NM394.<BR>7) NM394 strongly inhibited the supercoiling activities of DNA gyrase purified fromStaphylococcus aureus, Escherichia coliandP. aeruginosa.
