Abstract

吉野川水系の天然アユ集団の遡上状況と遺伝的多様性を評価するため, 遺伝マーカーを用い, 1997年, 1998年2年間に採集した合計18標本群を供試魚とし, 本調査を実施した。吉野川水系6地点からの10標本群および両側回遊型稚魚2標本群, 同河川に放流された放流用琵琶湖産種苗2標本群, 人工海産種苗4標本群について, マイクロサテライトDNAおよびアイソザイム多型を検出した。その結果, 吉野川産天然遡上アユは在来の海系アユの遺伝的基本特性を備え, 遺伝的多様性は十分高いことがわかった。陸封型 (琵琶湖産アユ) と両側回遊型 (海産アユ) の混合率から吉野川の天然遡上アユは池田ダムを越えて遡上していることが確認された。また, 吉野川へ放流された人工海産種苗の一部には遺伝的多様性が著しく低下した集団が認められた。

Using microsatellite DNA and isozymes genetic markers, we assayed allelic components of 18 sample lots of ayu consisting of the landlocked form juveniles collected from Biwa Lake (L1 and L2), the amphidromous form juveniles from Yoshino river and Katsuura river (A1 and A2), artificially propagated juveniles of amphidromous form (H1 to H4), and adult fish in question (Q1 to Q6-2) collected from lower to middle reaches of Yoshino river. Almost of all the ayu samples collected from Daiju dam to Ikeda dam (Q1-Q3) were identified as the native amphidromous form. The sample lots from Ikeda dam to the boundary of Tokushima prefecture (Q4-Q5) included fairly large amount of the landlocked form (37.1%-60.6%) . It was also ascertained that the amphidromous form was ascend to the upstream of the Ikeda dam through fishladder. The genetic variability of artificially propagated juveniles of amphidromous form being the seventh and eighth generation was low comparing with the sample lots from the wild population of landlocked and amphiromous form.
