16S rDNA塩基配列に基づいたキンギョ腸内細菌の同定 Identification of Goldfish Intestinal Bacteria by 16S Ribosomal DNA Sequence Analysis

Abstract

キンギョ, <I>Carassius auratus</I>の腸管内容物から分離した腸内細菌の16SrDNA塩基配列を調べ, 分子系統解析を行うとともに, 表現形質に基づく従来法の同定結果と比較した。キンギョ5尾から培養法により411菌株の好気性, 通性および偏性嫌気性細菌を分離し, 表現形質に基づいて同定した結果, これらは<I>Aeyomonas, Bacteroidaceae</I>, coryneforms, <I>Enterobacteriaceae, Pseudomonas</I>および<I>Vibrio</I>と同定された。次にこれらの分類群の代表菌46株について16SrDNA塩基配列を決定し, 系統分類学的検討を行った結果, これは<I>Aeromonas hydrophila, A.veronii</I>, CFB group, <I>Citrobacter freundii</I>, Fusobacteria, <I>Shewanella</I> sp., <I>Vagococcus</I> sp.および<I>Vibrio</I> sp.と同定された。種レベルで同定できた菌株は12株 (26.1%) にすぎず, 残りの34株 (73.9%) は新種または新属であることが強く示唆された。以上の結果から, 16SrDNA塩基配列 (約500bp) による同定法は識別能も高く, 魚類腸内細菌の同定に極めて有効であることが判明した。

In this study, taxonomic status of intestinal bacteria isolated from goldfish, <I>Carassius auratus</I>, was examined based on both phenotypic properties and 16S rDNA sequence. A total of 411 aerobic and anaerobic bacteria were isolated from intestinal contents of five goldfish specimens by the culture technique, and classified into six taxa, based on phenotypic properties: <I>Aeromonas, Bacteroidaceae</I>, coryneforms, <I>Enterobacteriaceae, Pseudomonas</I> and <I>Vibrio</I>. In addition, the 16S rDNAs amplified from 46 representative isolates were sequenced and their phylogenetic positions were analyzed. As a result, they were classified into eight taxa, including <I>Aeromonas hydrophila, A. veronii</I>, CFB group, <I>Citrobacter freundii</I>, Fusobacteria, <I>Shewanella</I> sp., <I>Vagococcus</I> sp. and <I>Vibrio</I> sp. on the basis of 16S rDNA sequences. These results mean that as many as 73.9% of tested isolates could not be identified as known species, suggesting that they correspond to new species or genera, which are composed of five taxa according to phylogentic analysis. Moreover, the identification method based on the first ca. 500 bp-sequence of 16S rDNA was proven to be adaptable for the further study of fish intestinal bacteria.
