ヤマブシ茸子実体のβ-D-Glucan Structural Analysis of the β-D-Glucan from the Fruit-body of Hericium erinaceum

Abstract

色調の異なる2種類のヤマブシ茸子実体(AおよびB)から脂溶性成分および冷水可溶性成分を除去し,その残渣をオートクレープ処理して熱水抽出粗多糖を得た.蛋白質を硫安沈澱で取り除いた後,DEAEsephadex chromatographyに供し精製多糖(AF2S-2およびBF2S-S)を得た.メチル化分析,スミス分解,酵素分解の結果から,これら多糖は共にβ-D-glucoseが(1→6)結合で主鎖を成し,(1→3)で枝分れした多糖であり,側鎖の鎖長は1~2であることが明らかとなった.また,重量平均分子量はそれぞれ13,000(AF2S-2)および22,000(BF2S-2)であった.

The polysaccharide (AF2S-2 and BF2S-2) was isolated from two types (A and B) of fruit-body of Hericium erinaceum via hot water extraction, fractional precipitation by ammonium sulfate, and DEAE-sephadex A-50 column chromatography. It was concluded from the results of the methylation study and Smith degradation that the polysaccharide was composed of a backbone of β-(1→6) -linked D-glucopyranosyl residues, and had β-D-glucopyranosyl and β-laminarabiosyl groups joined through 0-3 of n-glucose of the backbone. The structure was supported by the results of <SUP>1</SUP>H- and <SUP>13</SUP>C-NMR analyses and of enzymic hydrolysis of the polysaccharide. The molecular weight was estimated by gelfiltration to be 13, 000 (AF2S-2) and 22, 000 (BF2S-2).
