リゾチームによるN-Acetylglucosaminyl-Maltosyl-Cyclodextrinの合成 Synthesis of N-Acetylglucosaminyl-Maltosyl-Cyclodextrin by Lysozyme

Abstract

リゾチームの糖転移作用を利用して,N-acetylchito oligosaccharidesと分岐サイクロデキストリン(CD)からN-acetylglucosamineを結合したヘテロ分岐CDを合成した.N-acetylchitooligosaccharidesとmaltosyl-βCDからの転移物はN-acetylglucosamineがmaltosy1-βCDの分岐側鎖の非還元末端のグルコシル基にβ-1,3結合した,6-O-α-(3<SUP>2</SUP>-O-β-D-N-acetylglu cosaminyl-)maltosy1-βCD、と同定レた.リゾチームによるN-acetylglucosaminyl-maltosyl-βCD合成の60℃ における最適pHおよびpH4。5における最適温度はそれぞれ,3.5-6.0,60-70℃ であった.リゾチームを用いてN-acetylglucosamineをmaltosyl-βCD,およびgalactosyl-βCDと非分岐CDに結合させることはできなかった.

Hetero branched CDs linked by N-acetylglucosamine were synthesized from a mixture of N-acetylchitooligosaccharides and branched CDs by transglycosylation of lysozyme. A transfer product from N-acetylchitooligosaccharides and maltosyl-βCD was identified as 6-O-α-(3<SUP>2</SUP>-O-β-b-N-acetylglucosaminyl-) maltosylβCD. The optimum pH at 60°C and the optimum temperature at pH. 4.5 to produce N-Getylglucosaminyl-maltosyl-RCD by lysozyme were pH 3.5-6.0 and 60-70°C, respectively. Lysq zyme was unable to link N-acetylglucosamine to mannosyl-CD, galactosyl-CD and directly to CD rings.
