Abstract

雄豚9頭の精液を適宜使用して Percoll により不連続密度勾配層を作成し, X及びY精子の分別実験を行った。<br>すなわち, Percoll 上層部に精子浮遊液を積層し, 遠心分離を行い, 下層部に集まった精子と, その上の層に集まった精子を別々に吸引収集し, 精子数, 活力等の検査を行った。上下層精子とも収集精子数, 活力に若干の低下が認められたが特筆すべき影響はみられなかった。<br>ついで, これら両層由来の精子は30数頭の発情豚に対して人工授精された。<br>その結果, 受胎率, 産子数等は無処置の繁殖豚の受胎率や産子数と比べ特に差は認められなかった。<br>なお, 新生子の性比は, 上層部由来の精子で授精されたものと, 下層部由来の精子で授精されたものとの間に有意差はみられなかった。

Swine X and Y boaring spermatozoa from 9 boars of Duroc, Large Yorkshire and Berkshire, were separated by Percoll density gradient centrifugation.<br>A stock solution of Percoll was diluted with 0.15M-NaCl of the prescribed amount into 40-80% (7 layers). The spermatozoa were washed and loaded onto discontinuous gradients, and centrifugated at 1500rpm for 15min at room temperature.<br>The upper and bottom layer fractions only were separately collected and stored at 18°C. The concentration and motility of the spermatozoa were examined, but no influence was observed.<br>Twenty two sows and 12 sows were separately inseminated with the spermatozoa of the upper and bottom layer fractions, respectively. The number of offspring was the same as under normal circumstances and the sex ratio was not altered. Thus, the present procedure showed to be ineffective.
