Abstract

保存日数の経過に伴う精子の運動性, 生存率, 先体の形態および受精能との相互関係について検討した。濃厚部精液を Modena で4-5倍に希釈して10℃で保存した。精子の運動性は Motility Analyzer を用いて, Motile および Progressive を保存1, 7, 14, 21および28日目に解析した。精子の生存率および先体の形態の観察にはトリプルステイン法を用いた。また, 保存精子の受精能を体外受精法による精子侵入率で調べた。<br>保存精子の運動性は保存28日目においても高い運動性が維持され, 精子生存率も採取日の値に対して有意な変化は認あられなかった。しかしながら, 正常先体率は保存7日目以降急激に低下した (P<0.05)。保存精子の受精能の指標となる精子侵入率は, 正常先体率の低下に伴い, 保存14日目以降低下した。

This experiment was carried out to clarify various changes of the motility, acrosomal integrity and fertiliy of boar spermatozoa during liquid storage of 28 days at 10°C.<br>The sperm rich fraction was diluted to 1×10<sup>8</sup> spermatozoa/m<i>l</i> with Modena extender and stored at 10°C for 28 days. The motility and progressive of spermatozoa were analyzed by Motility Analyzer. The viability and acrosomal integrity were evaluated by Triple-Stain technique. The fertilizing ability of stored spermatozoa was evaluated by using in vitro fertilization technique.<br>The viability of spermatozoa was maintained in a good state for up to 28 days, while the spermatozoa with normal acrosome decreased after 7 days of storage, and <i>in vitro</i> fertilization rates decreased after 14 days of storage (p<0.05).<br>These results suggest that the boar semen diluted with Modena extender will be practically useful for the insemination until 7 days of storage at 10°C.
