Abstract

<p>We overview the recent development of single-molecule measurement based on light microscopy and spectroscopy. A key advantage of single-molecule measurement over ensemble measurement is distinct in revealing characteristics of individual molecules that are hidden under ensembles. Several topics are presented in light of this advantage. Also, another advantage of single-molecule measurement is emphasized in an application of chemical analysis, particularly in single-molecule DNA sequencing. Finally, special attention is paid to the use of surface-enhanced Raman spectroscopy, which is free from labeling target analytes, as a key tool of single-molecule chemical analysis.</p>
