Lymphocytes separated from thymoma (TLY) of myasthenia gravis (MG) patients were examined for their abilities to develop natural killer (NK) and lymphokine-activated killer (LAK) activities when stimulated by interleukin 2 (IL-2) and their phenotypic changes during activation with IL-2. Lymphocytes separated from non-thymomatous tissues of thymuses containing thymomas (NTLY) and peripheral blood lymphocytes (BLY) of MG patients were also examined for their abilities to develop NK and LAK activities and their phenotypic changes during activation. TLY of all MG patients developed LAK activity in response to incubation with recombinant IL-2 for 4 days. TLY of six of these seven patients also developed cytolytic activity against K 562 target cells. BLY of all of the patientts developed cytolytic activity against K 562 and LAK activity when incubated for 4 days with IL-2. The proportions of CD rcells and CD 25+ cells of TLY and NTLY of the MG patients increased significantly during incubation with IL-2 (P <0.05, P P<0.01, P<0.05). Although there was no significant phenotypic difference between TLY and NTLY of MG patients. LAK activity developed by TLY was significantly higher than that developed by NTLY (P<0.05).<BR>These results indicate that the TLY of MG patients differs functionally from the NTLY of MG patiens.
