Foreign Gene Expression in the Gonads of Chimaeric Chicken Embryos by Transfer of Primordial Germ Cells Transfected in vitro by Lipofection for 24 Hours

Abstract

Primordial germ cells isolated from the blood of early chicken embryos were transfected by lipofection for 24h and transferred to partially sterilised recipient embryos. The manipulated embryos were cultured for a further 3 days and the expression of foreign DNA (lacZ gene) in the gonads of the recipient embryos was examined. Expression of the lacZ gene was observed in the gonadal primordial germ cells of chimaeric embryos, suggesting that PGCs transfected in vitro by lipofection for 24h are still capable of migration to the germinal ridges of recipient embryos.
