Vas deferens strips of stroke-prone spontaneously hypertensive (SHRSP) and normotensive Wistar Kyoto (WKY) rats were tested for the sensitivity, contractile response and neuronal uptake ability in the response to norepinephrine (NE). Dose-response curves were determined before and after the inactivation of neuronal uptake system with cocaine. Sensitivity and contractile response to NE in the epididymal and prostatic ends of vas deferens of SHRSP were greater than those of WKY. After the addition of cocaine, all the preparations of SHRSP and WKY showed an increased sensitivity without a potentiation of maximum tension of NE. Cocaine also produced a leftward shift on the responses to K^+ and methoxamine in the SHRSP but not in the WKY. The increases in sensitivity to NE were less in the SHRSP, indicating that the neuronal uptake ability was reduced in the SHRSP. NE and methoxamine responses in the presence of cocaine were still greater in the epididymal end of SHRSP. These results suggest that the increases in sensitivity and contractile response to NE in the epididymal and prostatic ends of SHRSP contribute to both the decrease in neuronal uptake and the increase in NE response of smooth muscle cells. In addition, the epididymal ends in both strains of rats were greater than the prostatic ones in the response to NE and methoxamine. The data also suggest that the regional differences are probably related to other natures rather than the neuronal uptake mechanism.
