The purpose of this study was to investigatewhether lidocaine could be transported throughexcised rat skin in vitro using alternating current(AC). In addition, the relationships between factorssuch as voltage and frequency, and transportedlidocaine concentration were studied using the invitro model. A pair of platinum plate electrodes wasinstalled at opposite ends of two cylindrical glasscells in parallel to the full-thickness rat skin. Thedonor compartment was filled with 10 % lidocainehydrochloride, and the receptor compartmentwith Ringer solution. A sinusoidal wave wasapplied between the electrodes at 5 kinds of constantvoltages at 1 kHz and at 4 kinds of frequenciesat 20 volts. Our experimental system wassuccessfully used to quantify the concentration oftransported lidocaine induced by AC application.The applied sinusoidal waves evoked the transportof lidocaine through the rat skin at every voltageand frequency protocol. Our results suggest thatthe applied electric voltage and frequency affectthe movement of the lidocaine ions. We concludethat the transdermal delivery of lidocaine by ACiontophoresis has a possibility to use for localanesthesia and the pain management of the skin.
