Decalcified adult bone and dentin sections were hybridized with cRNA probe of bone matrix proteins such as osteonectin, osteopontin, osteocalcin and matrix Gla protein labeled by digoxigenin-11-UTP. The system of in situ hybridization has a higyer resolution, makeing it easier to identify the cell types which an expressing the gene, than using 35S or 32P labeled probes. Using the system, the localization of the mRNA for bone matrix protein such as osteonectin (Osn), osteopontin (Osp) and matrix Gla protein (MGP) was examined. Decalcified femur and dentin of neonatal to 40-week old mice were used for examination. Osn and Osc mRNAs were localized in osteoblast of bone and odontoblast in dentin. Osp mRNA was localized in osteoblast but no apparent signal was observed in dentin. MGP mRNA was detected in hypertrophic chondrocytes. Further identification of thecell types which expressed the gene was carried out. These results indicated the usefulness of this system to investigate the relationship of the positional information and gene expression.
