Effect of isometric exercise on changes in contractile function during recovery after hindlimb suspension (HS) was investigated in the soleus muscles of adult (4-5 months) and old (20-21 months) rats. The isometric contractile properties were measured in situ. In both adult and old rats, soleus wet weight and maximum tetanic tension significantly decreased during 3 weeks of HS and returned to about 80% of control level during 3 weeks of recovery after HS. Effect of isometric exercise (stationary support on a grid at 80° with a load equal to 50% of body weight attached to the tail for 30 min per day, 3 or 6 days per week) on the recovery of muscle weight and maximum tetanic tension was not found. Contraction (CT) and half-relaxation (HRT) times of the isometric twitch decreased during. HS and increased during recovery in both adult and old rats. However, the percentage of changes in the CT and HRT during HS and recovery was smaller in old rat compared to in adult rat. This result indicates that response to altered muscle activity declines in old age. The isometric exercise (6 days per week) accelerated recovery of CT and HRT in old rats. It is suggested from present study that an additional muscle activity during recovery after reduced-use play an important role for improvement of muscle function in old age.
