Abstract

ビタミンEの周産期において果たす役割について知る目的で, 血小板凝集を中心に検討し以下の成績を得た. 1. plasma中ビタミンE(α-tocopherol)濃度(μg/ml)は, 非妊娠婦人:6.65, 妊娠婦人(37~40週):15.5であり妊娠経過とともに増加を認めた. また, 一方, 血小板中ビタミンE濃度(μg/g protein)も非妊娠婦人:99, 妊娠婦人(37~40週):244で妊娠経過とともに増加を認めた. 2. ビタミンEには, invitroで, ADP(adenosine diphosphate), PAF(plateletactivating factor)惹起血小板凝集の阻止活性を認め, 500μg/mlの濃度で, ADPおよびPAFの血小板凝集をほぼ100%阻止した. 3. ヒト胎盤絨毛上皮刷子縁膜小胞には著明な血小板凝集阻止活性が存在しており, 20μg/ml(終蛋白濃度)では, ADPおよびPAFによる血小板凝集をほぼ100%阻止した. 4. ヒト胎盤絨毛上皮刷子縁膜小胞存在下では, ビタミンEの血小板凝集阻止作用は, 著明に増強された. 以上の成績より, 母体血小板中ビタミンE濃度は, 妊娠経過とともに著明に増加し, 胎盤絨毛組織内には, 比較的高濃度に, ビタミンEが存在することが判明した. また, ビタミンEには血小板凝集阻止活性があり, 胎盤内の微小循環の維持への関与も示された.

To clarify the role of vitamin E(α-tocopherol) during pregnancy, we mainly investigated the relation between platelet function and vitamin E and obtained the following results. 1. The concentration of vitamin E(α-tocopherol) in plasma and platelet increased gradually during pregnancy. The concentration of vitamin E in plasma was 6.65μg/ml in non pregnant women and 15.5μg/ml in full term pregnant women. And the concentration of vitamin E in platelet was 99μg/g protein in non pregnant women and 244μg/g protein in full term pregnant women. 2. Vitamin E(α-tocopherol) inhibited platelet aggregation induced by ADP(adenosine diphosphate) and PAF(platelet activating factor). At a concentration of 500μg/ml vitamin E almost combletely inhibited platelet aggregation induced by ADP and PAF. 3. There exists a strong platelet aggregation inhibiting activity in human placental brush border membrane vesicles. At a protein concentration of 20μg/ml the brush border membrane vesicles almost completely inhibited platelet aggregation induced by ADP and PAF. 4. With the co-existence of the brush border membrane vesicles, the platelet aggregation inhibiting activity of vitamin E(α-tocopherol) increased prominently. These results indicate that vitamin E plays an important role in the regulation of platelet function and might contribute to the maintenance of placental microcirculation by inhibiting platetelet aggregation.
