Abstract

アトピー患者白血球および受動的感作ヒト肺切片からのアナフィラキシー性histamineおよびleukotriene (LT) 遊離に及ぼすtranilastの影響について検討した.1.10^<-5>g/mlのtranilastの反応惹起30min前処理は, 末梢血白血球からのhistamine遊離を抑制しなかったが, peptide LTsの遊離を抑制した.2.10^<-5>および10^<-4>g/mlのtranilastの反応惹起30min前処理は, 感作ヒト肺切片からのhistamine遊離のみならず, LTB_4およびpeptide LTsの遊離をも抑制した.以上の成績から, tranilastの抗喘息効果の一端は, histamineおよびLT遊離を抑制する機構によることが示唆された.

The effect of tranilast on anaphylactic histamine and leukotriene (LT) release from leukocytes from atopic patients and passively sensitized human lung fragments were examined. 1.Treatment with tranilast (10^<-5> g/ml) for 30 min prior to antigen challenge moderately inhibited peptide LT release from human atopic leukocytes but did not prevent histamine release. 2.Treatment with tranilast (10^<-5> and 10^<-4> g/ml) for 30 min prior to antigen challenge inhibited the release of not only LTB_4, LTC_4, LTD_4 and LTE_4 but also histamine from passively sensitized human lung fragments. From these results, it was suggested that the suppression of LT as well as histamine release contributed to the antiasthmatic effect of tranilast.
