The transport capacity and direction of indole-acetic acid (IAA) in roots were analyzed in large carrot of horticultural cultivar, and furthermore extractable and diffusible IAA were determined. The root were divided into tissues (epidermis, cortex, phloem, xylem and pith), which were further divided into upper (base), middle and lower (distal) parts. As to the analysis of IAA transport, the end of the section was applied radio-labeled IAA and after 24 hours radioactivity of the other end of the section was determined, and the difference between basipetal and acropetal directions was designated as polar transport capacity. In the long axis of the root, the basipetal direction of the polar transport capacity was confirmed in the epidermis and cortex. In the phloem, xylem and pith, the difference between basipetal and acropetal directions was not significant. In the short axis of the root, there was no significant difference between outward and inward direction. As to extractable IAA, 13C6-IAA as an internal standard was added, and IAA was determined with LC-MS. The concentrations of extractable IAA were generally higher in upper parts in each tissue, especially in the epidermis and cortex. As to diffusible IAA, the end of the section was soaked in distilled water, which was determined with LC-MS as the extractable IAA. The concentrations of diffusible IAA were higher in upper parts in phloem, xylem and pith tissues. In this reports, the basipetal stream of IAA through the epidermis and cortex was confirmed.
