THIOCTAMIDEの生体内変化について : (I)ウサギにTHIOCTAMIDEを投与したさいの血中THIOCTIC ACID活性物質の検討 THE CONVERSION OF THIOCTAMIDE TO THIOCTIC ACID IN BIOLOGICAL SYSTEM : (I) STUDIES ON THE THIOCTIC ACID ACTIVE SUBSTANCES IN RABBITS SERUM AFTER THE ADMINISTRATION OF THIOCTAMIDE
When thioctamide was administrated per os in men or rabbits, the level of the serum thioctic acid activity was immediately elevated and then successively recovered. Comparable results were obtained in the case of thioctic acid administration in men. In bioautographic analysis of rabbit serum after the administration of thioctamide, thioctamide was unexpectedly not detected, but a large amount of free thioctic acid was found with a small amount of its sulfoxide. It therefore appeared that thioctamide was converted to thioctic acid in vivo. In the experiment in vitro, this conversion was also observed in rabbit blood and other tissue homogenates. The conversion activity of each tissue preparation was lost by heat treatment. It would be considerable that the conversion of thioctamide to free acid is a reaction catalysed by enzyme.
