A simple, rapid and reproducible method for determination of rifampicin in serum by high-performance liquid chromatography has been developed. Only 20 μl of serum is required. Serum proteins are precipitated with 100 μl of methanol and rifampicin is dissolved into the methanol solution. After the centrifugation, 90 μl of the supernatant is injected into the chromatograph equipped with a Model U6K injector, a 3.9 mm × 30 cm μBondapak C18 column, a Model 440 UV detector set at 0.01 AUFS (340 nun) and a 6000 A solvent delivery system (Waters Assoc. Inc., ). The flow rate of the mobile phase of 38 % acetonitrile solution containing 0.01 M sodium acetate (pH 7.0) is 1.5 ml/min.<BR>The retention time for rifampicin is 9 min. The amount of rifampicin is determined by measuring peak heights. The limit of detection is 0.5μg/ml of serum. Within-run and day-to-day reproducibility for 8.0 μg/ml are ± 1.0% (n =10) and ± 1.5% (n =10) respectively. The proposed method can be completed in less than 13 min and had been applied for the serum of volunteers taking rifampicin.
