Abstract

DNA分子マーカーを利用した<I>Acacia mangium</I>と<I>A. auriculiformis</I>の種間雑種の鑑定法を確立した。RAPD分析によりそれぞれの種に特異的なDNAフラグメントをスクリーニングし,その塩基配列情報を基に42個のSCAR(sequence characterized amplified region)マーカーを設計した。これらのマーカーの中に,高い種特異性を有し,異なる表現型(フラグメント長)を示す2個の共優性マーカーが存在した。この2マーカーを利用することにより雑種個体を高い信頼性で鑑定することが可能になった。

We constructed a diagnostic system of the interspecific hybrids between <I>A. mangium</I> and <I>A. auriculiformis</I> using DNA molecular markers. The species-specific RAPD (random amplified polymorphic DNA) fragments were selected in each species, and 42 SCAR markers were designed based on the sequence data of these fragments. Two codominant markers were detected in the SCAR markers. They are highly species-specific and show the phenotypes with different fragment length between the two species. These markers make it possible to reliably identify putative interspecific hybrids.
