In order to analyze fiber types of skeletal muscles on tissue sections, we raised antibodies to myosin of the chick pectoral muscle and its S-1 fragments and these to myosin of the chick cardiac muscle and its S-1 fragments in rabbits. Similar to the enzyme histochemistry for myosin ATP ase at different pHs on serial frozen sections, the antibodies successfully distinguished type I and type II fibers: the antipectoral myosin/S-1 cross-reacted with type II fibers and the anti-cardiac myosin/S-1 cross-reacted with type I. Their specificities were also determined by Western blotting analysis. These antibodies were able to discriminate muscle fiber types even on large-sized formalin-fixed paraffin sections by immunoperoxidase techniques. Using this method, we found coexpression of plural myosin isoforms within a single muscle fiber of human lingual muscle. The immunohistochemical method using these specific antibodies against myosin isoforms was shown to have considerable advantages over the conventional enzyme histochemistry that was only possible on frozen sections.
