Most of the α-amylase in germinating Phaseolus vidissimus Ten. seeds is found in the cotyledon. α-Amylase was isolated in a highly purified form. The molecular weight was estimated to be 59,000 from the mobility on SDS-polyacrylamide gel electrophoresis. The enzyme readily hydrolyzed amylopectin, amylose, glycogen and soluble starch, and maltotriose very slightly. At the hydrolysis degree of about 18% of soluble starch, at which the iodine color disappeared, oligosaccharides larger than maltotetraose largely accumulate in the reaction mixture, and glucose, maltose and maltotriose a little.
