Abstract

DDTおよびdieldrinをマウスに経口または静脈内投与すると, DDTでは振戦, dieldrinでは強直性および間代性痙攣がみられ, 脳室内投与によっても質的に類似した症状が引き起こされた. 静脈内注射された両剤は, 神経切断後の坐骨神経-腓腹筋系に直接作用を示さなかったことより, 中枢神経系に由来する作用と思われた. DDTの振戦は抗ドーパミン薬で悪化したが, 抗ムスカリン薬は影響しなかった. dieldrin痙攣は抗ムスカリン薬または抗ドーパミン薬で軽減され, 抗アドレナリン薬または抗GABA薬で悪化された, このようなDDTとdieldrinとの相違から, 両剤が異なる神経路を通じて神経毒性を発現させていることが示唆された.

Neurotoxic effects of clofenotane (DDT) and dieldrin were analyzed pharmacologically in mice. Oral (p. o.) or intravenous (i. v.) administration of DDT caused the tremor, while dieldrin caused the tonic or clonic convulsion. Intracerebroventricular (i. c. v.) injection of both agents also caused the neurotoxic symptoms which were qualitatively similar to those shown after p. o. or i. v. administration. These effects may be central nervous system in origin since i. v. injection of either insecticides had no direct effect on sciatic nerve-gastrocnemius muscle system after the nerve had been sectioned. The tremor due to DDT was aggravated by dopaminergic antagonist but unaffected by muscarinic antagonist. The convulsion due to dieldrin was alleviated by muscarinic or dopaminergic antagonist but aggravated by adrenergic or GABA ergic antagonist. These differences suggest that DDT and dieldrin develop their neurotoxic symptoms through different neural pathway(s).
