Removal and sedation of hyperalgesia as well as treatment of tooth pain are the two of the major aim of dental treatments. With the use of a few compounds, such as eugenol or other phenol derivatives, hyperalgesia can be controlled. However, these compounds have an adverse effect for the dental pain control. Recent developments of molecular biology reveal new insights for the molecular mechanisms of nociception. In this review, we propose several possible targets for dental sedatives, such as bradykinin receptors, prostaglandin receptors, capsaicin receptors, ATP reeptors, voltage-dependent Na channels and voltage-dependent Ca channels. As activation of these receptors and ion channels promotes pain and inflammation, any block or inhibition of these targets would suppress the pain. Thus, the development of new drugs targeted to these sites may lead to the full control of the tooth pain.
