Cj57 protein is a pollen allergen from Camellia japonica with a molecular weight of 57kDa and an isoelectric point of pI 5.4. To study the function of Cj57 during pollen germination, the properties of Cj57 were examined using a specific antibody directed against it. Cj57 was detected in different pollen grains, but was not present in the petals and sepals. The level of Cj57 in the cytoplasm was very low in mature quiescent pollen grains, but it increased rapidly after pollen hydration: the level also decreased after incubation of the pollen grains for 10min. During further expansion, germination and subsequent tube growth, Cj57 was scarcely detected. Immunoelectron microscopy confirmed these characteristic changes in the level of Cj57 during pollen growth. Cj57 was also analyzed by mass spectrometry and a sequence homology search indicated that it is related to heat shock proteins. These results suggest that Cj57 may be involved in the early stage of pollen germination, for example, the regulation of water stress after pollen hydration.
