Abstract

According to the National Nutrition Survey in Japan, dietary fat intakes have been increasing for the past several decades. In order to understand how Japanese women take in fat during pregnancy, we first conducted a survey on the dietary habits of 156 normal pregnant women using diet history questionnaires. Our survey data showed that there was a significant positive association between dietary caloric intake and fat consumption, and that the daily saturated fat intake is greater than recommended level in Japan. Next, to investigate the effects of maternal high fat diet (HFD) on maternal insulin resistance, animal experiments focusing on the inflammation of adipose tissue were conducted, yielding the following findings: i) The HFD dams exhibited impaired glucose tolerance and increased insulin resistance; and ii) M1 (pro-inflammatory) macrophage infiltration into subcutaneous and parametrial adipose tissue was increased in the HFD dams. These findings suggest that maternal HFD leads to increased maternal insulin resistance via adipose tissue inflammation. Finally, to observe the effects of maternal HFD on blood pressure and glucose and lipid metabolism in adult offspring, another set of animal experiments was conducted, yielding the following results: i) The offspring of HFD dams (HDF offspring) showed hypertension and impaired glucose and lipid metabolism, even without the presence of obesity: ii) Serum adiponectin levels in the HFD offspring decreased, and the HFD offspring had enlarged mesenteric fat cells; and iii) In mesenteric adipose tissue of the HFD offspring, TNF-a and CDllc mRNA levels were increased. These results indicate that maternal HFD leads to a metabolic syndrome-like phenotype in adult offspring via mesenteric adipose tissue inflammation. Taken together, not only does maternal high-fat diet increase maternal insulin resistance, but it also leads to a metabolic syndrome-like phenotype in the adult mouse offspring.
